Top 100 most followed tweeters here :http://twittercounter.com/pages/100, which is a cool resource if you think your targets might be into Lady Gaga or Justin Beiber.

Probably best to look for big influencers in your own market space though.

I find it pays to think laterally (what parallel's are there with what you're doing) and to go for quality over quantity and to actually interact with people. There's an awful lot of following for the sake of it, which never turns into meaningful interaction.
